ExoSAP-IT PCR Product Cleanup
ExoSAP-IT® is a single-step PCR cleanup product available from USB Affymetrix.
ExoSAP-IT® has several advantages over column purification including:
Time saving
- Single-step PCR cleanup
- Easy to clean up multiple samples at the same time
- Eliminates hassle of columns
Effective
- Enzymatic cleanup removes primers and dNTPs
- Zero sample loss
Better value
- Easier workflow and lower cost per prep than columns
Description:
The ExoSAP-IT method is a unique, one-step enzymatic cleanup of PCR products that eliminates unincorporated primers and dNTPs so they do not interfere with downstream sequencing or SNP analysis. The ExoSAP-IT cleanup reagent, based on Exonuclease I and Shrimp Alkaline Phosphatase, is added directly to the PCR product to degrade primers and dephosphorylate dNTPs that were not consumed in the reaction (Figure 1). Since the ExoSAP-IT reagent is active in commonly used PCR buffers, no buffer exchange is required. Treatment is carried out for 15 minutes at 37°C and is followed by a 15-minute incubation period at 80°C to completely inactivate both enzymes. PCR products are then immediately ready for downstream sequencing reactions without any additional manipulation.

Fluorescent sequencing results of a 100 bp pUC18 PCR fragment sequenced with a -20 Fwd primer using fluorescent sequencing reagents. PCR cleanup performed with: (a) ExoSAP-IT reagent; (b) a column designed for PCR cleanup. Base miscalls in (b) are due to inherently low yields of short PCR products when using columns.

NGAL – Early Diagnosis of Acute Kidney Injury
In recent years NGAL has been shown to be perhaps the earliest biomarker to respond to acute kidney injury. After such injury, NGAL levels rise quickly and significantly in both urine and blood, showing a much wider response range than many other proposed biomarkers.
BioPorto Diagnostics was the first to market NGAL antibodies and assays. Their animal NGAL ELISA kits are intended for use in research and preclinical studies to detect rises in NGAL concentrations in urine and blood and hence the extent of renal injury, including nephrotoxic injury.
The applications of BioPorto NGAL ELISA kits range from basic research and drug discovery to preclinical toxicity testing. BioPorto also offers human NGAL ELISA kits which can be used in clinical studies, to detect nephrotoxicity in the relevant phases of clinical trials.

Human p450 cDNA Plate Array
Cytochrome P450 is a multi-gene family of enzymes, which play a central role in the oxidative metabolism of a wide range of xenobiotics, including anticancer drugs. Human CYP genes encode fifty-seven P450 proteins with numerous homologue variants in different tissues. They are highly differentially expressed in response to a variety of environmental chemicals and pharmaceuticals.
Signosis have developed a P450 cDNA Plate Array for profiling the expression of a panel of 22 of these CYP genes.
Human p450 cDNA Plate Array, Catalogue Number: AP-0202

Analysis of expression of P450 genes. 2ug of RNAs from MCF7, HL-60, HeLa and 293 cell lines were subjected to the P450 cDNA plate assay.
